SS-31 (Elamipretide): The Mitochondria Peptide Explained Simply
If you spend time in longevity threads you have probably seen SS-31 come up, usually right next to the phrase "targets your mitochondria." That sounds impressive and also kind of vague. So let me break down what SS-31 is, what it actually does at the cellular level, and where the research really stands, without the usual breathless spin.

SS-31 is one of the more legitimately studied peptides in this niche, which is refreshing. It even has a real drug name.
What SS-31 is and what “mitochondria-targeting” means
SS-31 is a small peptide, just four amino acids, also known as elamipretide (and sometimes MTP-131). The "SS" comes from the two scientists who developed it, Szeto and Schiller.
The interesting part is where it goes. Most molecules float around the cell more or less at random. SS-31 concentrates in the inner membrane of your mitochondria, specifically binding to a lipid called cardiolipin. That membrane is where your cells make most of their energy. When it gets damaged or leaky, energy production drops and harmful byproducts go up.
The theory is that by parking itself in that membrane, SS-31 helps it work more efficiently and cuts down on the cellular equivalent of exhaust fumes. That is why you see it lumped in with other "mitochondrial health" compounds. It is not a stimulant and it is not a hormone. It is a membrane-stabilizer, basically.
What the research on SS-31 actually shows
Here is the part that separates SS-31 from a lot of the hype-peptides: it has been in real clinical trials in humans. Elamipretide has been studied for conditions tied to mitochondrial dysfunction, including a rare genetic disorder called Barth syndrome, as well as certain heart and eye conditions.
Elamipretide has been under FDA review for Barth syndrome, which is a big deal in a field where most compounds have only ever seen a mouse. That said, the road has been bumpy. Some trials hit their marks and some did not, and the approval process has not been a straight line. So "in clinical trials" does not automatically mean "proven to work for everything the internet claims."
And to be clear, outside of that specific review, SS-31 as a general research peptide is not FDA-approved for anti-aging, energy, or any of the longevity uses people are actually curious about. That gap between "studied for a rare disease" and "sold as a wellness peptide" is enormous, and worth keeping in mind.
Why the longevity crowd cares
The appeal is easy to understand. If aging is partly a story of mitochondria wearing out, a compound that stabilizes the exact membrane where energy is made sounds like it should help. It fits the theory beautifully.
People often research it in the same breath as MOTS-c and NAD+, treating the three as a "mitochondrial support" trio. I get the logic. I also want to gently point out that a clean mechanism and a genuine disease trial do not add up to "this will make a healthy 40-year-old feel 25 again." The evidence for that just isn't there yet.
